Cada rata fue inoculada inyectándole por vía intraperitoneal 0.1 mL de la suspensión sanguínea con 1x105 tripomastigotes sanguícolas de Trypanosoma cruzi (cepa I/PAS/VE/00/PLANALTO). Los parásitos fueron aislados de Panstrongylus geniculatus, naturalmente infectado y capturado en un área urbana del valle de Caracas, Venezuela y mantenidos en ratones NMRI. A los 10 días post-infección a estas ratas (GI) les fue determinado el ciclo estral y luego fueron apareadas con los machos en una relación 2 hembras/1 macho en cada jaula. Grupos de ratas vírgenes inoculadas con T. cruzi (GII), ratas sanas (GIII) y ratas preñadas (GIV) fueron usadas como controles. La infección por T. cruzi en las ratas de GI reveló los más altos niveles de parasitemias patentes con 12,2 ± 1.1 T. cruzi/mm3 de sangre a los 18, 24 y 34 días postinfección con 6, 12 y 20 días de gestación respectivamente, al compararla con el GII. Los títulos de anticuerpos específicos anti-T. cruzi fueron mayores en el grupo de ratas gestantes e infectadas con T. cruzi, en comparación con las vírgenes infectadas, representados por 1:512 y 1:2048 a los 19 y 20 días post-infección respectivamente. El 33 % de las muestras de líquido amniótico de las ratas del GI, desarrollaron formas flageladas de T. cruzi en el medio de cultivo NNN. El estudio histopatológico de los cortes de 6μ de espesor coloreados con hematoxilina y eosina del corazón fetal (CF) obtenidos de 2 fetos de ratas del GI con 34 días de infección, reveló nidos de amastigotes en el tejido cardíaco. En las placentas de las ratas del GI se observó una placentitis caracterizadapor intenso infiltrado inflamatorio de células mononucleares y polimorfonucleares con predominio de neutrófilos. Los ensayos inmunocitoquímicos con el doble marcaje Isotiocianato de fluoresceínaioduro de propídio y peroxidasa-anti-peroxidasa en los cortes de placenta, corazón fetal y cordón umbilical de las ratas del GI, mostraron parásitos y abundante antígeno de T. cruzi. Estos resultados demostraron la infección intrauterina fetal por T. cruzi confirmando la transmisión transplacentaria del parásito desde la madre infectada a su progenie. Posiblemente el mecanismo de transmisión congénita esté relacionado con la liberación de los tripomastigotes a través del trofoblasto, a la aspiración del líquido amniótico por los fetos o a la penetración activa de los parásitos a través de la piel y/o mucosas durante el período fetal(AU) aThis study was carried out with a group of 20 days old, 250-g female juvenile rats (Rattus inoculated, injecting it intraperitoneally with a 0.1- mL suspension of Blood with 1 x 105 tripomastigotes from the I/PAS/00/PLANALTO strain. The parasites were isolated from naturally infected Panstrongylus geniculatus from a street in an urban area in Caracas, Venezuela, and maintained in NMRI rats. At 10 days post-infection (pi), the rats were determined to have completed the estrual cycle and were then mated with males, with a ratio of 2 females and 1 male in each cage. Groups of virgin rats inoculated with T. cruzi (GII), healthy rats (GIII) and pregnant rats (GIV) were used as controls. Infection by T. cruzi in the GI rats revealed the highest levels of clear parasitemias with 12.2 ± 1.1 tripomastigotes/mm3 of blood at 18, 24 and 34 days (pi) with 6, 12 and 20 days from gestation, respectively, compared with GII. The titles of specific antibodies anti-T. cruzi were greater in the group of pregnant and infected rats in comparison with the infected unmated rats, represented by 1:512 and 1:2048 at 19 and 20 days (pi) respectively. 33% of the samples of amniotic fluid of the rats of GI developed flagellate forms of T. cruzi in NNN culture. Histopathological study of 6μm sections stained with hematoxilin-eosin of fetal hearts obtained from 2 rat fetuses revealed nests of amastigotes in the cardiac tissue. In the placentas of GI with 34 days of infection, we observed a placentitis characterized by intense inflammatory infiltration of the mononuclear and polymorphonuclear cells with neutrophyls predominant. The immunohistochemical assays using double marking with fluorescein isothiocyanatepropidium iodide and peroxidase-antiperoxidase in the sections of placenta, fetal hearths and umbilical cords showed parasites and abundant antigen of T. cruzi. These results demonstrate the fetal intrauterine infection by T. cruzi, confirming the transplacental transmission of the parasite from the infected mother to her offspring.
